I think the depletion of natural resources will be the big one. I also think though that most of the future challenges are increasingly interrelated and hard to separate from each other.
i believe that a conflict between them is unlikely now a days, those countries are still very dependent on each other economically, engage in an armed conflict provides only desavantages for both of them.
I think religious extremism and attempts to grab power by non-state actors will be the biggest challange because countries can resolve issues by diplomacy but handling non-state actors is very tough.
The quest of power might led us to a third world war. Other than this, the way young generation is being brought up, loss of moral values and ethics will be a huge problem. See, it keeps everything in place.
From the ratio of participants answering correctly challenges on this forum, I think that the greatest challenge facing the humanity, is an excess of mathematicians compared to a shortage of physicists:)

Ethics concerning robots if they ever become sentient. I'm actually kinda afraid of that one.

Yeah, I think that its getting very tense between USA and other nations. That could be a growing problem...
